What you need is a direct male descendant. In other words, you need an uninterrupted line of sons. You can't go "son, son, daughter, son" because women only carry female chromosomes, while men have both male and female. In other words, find a living male heir of on of Jame's sons that is still living and comes from an unbroken succession of male heirs and give him a Y-DNA test. On the maternal line, you can test any male or female who's maternal line traces back to Harriett. You can also have several other relatives take the test to get other branches of the family tree. Males are always best for testing because they can be tested for both paternal and maternal lineage. Hope this answers your question.
